Structure-Function Relationships in DNA Polymerases

Marc Delarue
CNRS, France/ Institut Pasteur
Structural Biology and Chemistry

The following subjects will be covered. Sequence analysis, multalignement and classification of DNA polymerases. A worked example in the study of structure-function relationships of a member of the pol X family: terminal desoxynucleotidyltransferase (TdT). Summary of what is known on the different conformational changes of the enzyme in the full catalytic cycle of nucleotide addition. Simulation of these transitions using simplified models (ENM): an attempt to understand the translocation step.
