Single-particle Reconstruction of the Ribosome -- The Struggle to Get Toward Atomic Resolution

The extraordinary amount of noise in EM low-dose images of molecules makes attainment of high resolution very difficult. The ribosome is on a steady course in the right direction, but the progress has been incremental. Among the challenges are the problems of conformational heterogeneity, the need for highly accurate CTF correction, and interpretation of density maps by flexible fitting. Recent progress in 3D imaging of the ribosome will be presented.
